Penghambatan Aktivitas Enzim Aldosa Reduktase dari Senyawa Aktif Daun Tanaman Kelor (Moringa Oleifera L.): Studi In-Silico

Di Indonesia, terdapat berbagai macam tumbuhan yang dapat digunakan sebagai obat tradisional, salah satunya yaitu daun tanaman kelor (Moringa oleifera L.) berpotensi antidiabetes. Proses penemuan memerlukan tahapan banyak dan waktu lama. Salah satu cara dalam mempercepat proses kandidat baru adalah dengan pendekatan secara komputasi docking molekular. Bahan penelitian protein 2HV5 pada enzim aldosa reduktase, ligan ZST, senyawa aktif marumoside A, quercertin, asam ferulat, kaempferol galat. Berdasarkan validasi metode molekuler, diperoleh nilai RMSD 0,8704 Amstrong, hasil best score dari masing-masing berikut: ZST (-126,7120); Pioglitazine (-120,4064); Sitagliptin (-120,056); Marumoside A (-93,78563); Quercertin (-80,9221); Asam Ferulat (-80,2762); Kaempferol (-80,09625); Galat (-72,6102), dimana semua memenuhi aturan Lipinski. Dapat disimpulkan bahwa kelima tersebut kurang potensial jika dibandingkan pioglitazine sitagliptin serta perlu dilanjutkan uji aktivitas in vitro di laboratorium.
